Designing gene drives to limit spillover to non-target populations

The prospect of utilizing CRISPR-based gene-drive technology for controlling populations has generated much excitement. However, the potential spillovers alleles from target population to non-target raised concerns. Here, using mathematical models, we investigate possibility limiting by designing differential-targeting gene drives, in which expected equilibrium allele frequencies are high but low population. We find that achieving differential targeting is possible with certain configurations parameters, but, most cases, only under relatively migration rates between populations. Under migration, a narrow region parameter space. Because fixation drive could severely disrupt ecosystems, outline ways avoid this outcome. apply our model two applications drives—field trials malaria-vector drives and control invasive species on islands. discuss theoretical predictions key requirements their practical implications.
